    Development of carbon slurry fuels for transportation (hybrid fuels, phase 2)

    Slurry fuels of various forms of solids in diesel fuel are developed and evaluated for their relative potential as fuel for diesel engines. Thirteen test fuels with different solids concentrations are formulated using eight different materials. A variety of properties are examined including ash content, sulfur content, particle size distribution, and rheological properties. Attempts are made to determine the effects of these variations on these fuel properties on injection, atomization, and combustion processes. The slurries are also tested in a single cylinder CLR engine in both direct injection and prechamber configurations. The data includes the normal performance parameters as well as heat release rates and emissions. The slurries perform very much like the baseline fuel. The combustion data indicate that a large fraction (90 percent or more) of the solids are burning in the engine. It appears that the prechamber engine configuration is more tolerant of the slurries than the direct injection configuration

    Intravital Multiphoton Microscopy with Fluorescent Bile Salts in Rats as an In Vivo Biomarker for Hepatobiliary Transport Inhibition

    The bile salt export pump (BSEP) is expressed at the canalicular domain of hepatocytes, where it mediates the elimination of monovalent bile salts into the bile. Inhibition of BSEP is considered a susceptibility factor for drug-induced liver injury that often goes undetected during nonclinical testing. Although in vitro assays exist for screening BSEP inhibition, a reliable and specific method for confirming Bsep inhibition in vivo would be a valuable follow up to a BSEP screening strategy, helping to put a translatable context around in vitro inhibition data, incorporating processes such as metabolism, protein binding, and other exposure properties that are lacking in most in vitro BSEP models. Here, we describe studies in which methods of quantitative intravital microscopy were used to identify dose-dependent effects of two known BSEP/Bsep inhibitors, 2-[4-[4-(butylcarbamoyl)-2-[(2,4-dichlorophenyl)sulfonylamino]phenoxy]-3-methoxyphenyl]acetic acid (AMG-009) and bosentan, on hepatocellular transport of the fluorescent bile salts cholylglycyl amidofluorescein and cholyl-lysyl-fluorescein in rats. Results of these studies demonstrate that the intravital microscopy approach is capable of detecting Bsep inhibition at drug doses well below those found to increase serum bile acid levels, and also indicate that basolateral efflux transporters play a significant role in preventing cytosolic accumulation of bile acids under conditions of Bsep inhibition in rats. Studies of this kind can both improve our understanding of exposures needed to inhibit Bsep in vivo and provide unique insights into drug effects in ways that can improve our ability interpret animal studies for the prediction of human drug hepatotoxicity

    It is demonstrated that, in the adiabatic approximation, non-Equatorial circular orbits of particles in the Kerr metric (i.e. orbits of constant Boyer-Lindquist radius) remain circular under the influence of gravitational radiation reaction.     Very long baseline astrometry of PSR J1012+5307 and its implications on alternative theories of gravity

    PSR J1012+5307, a millisecond pulsar in orbit with a helium white dwarf (WD), has been timed with high precision for about 25 years. One of the main objectives of this long-term timing is to use the large asymmetry in gravitational binding energy between the neutron star and the WD to test gravitational theories. Such tests, however, will be eventually limited by the accuracy of the distance to the pulsar. Here, we present VLBI (very long baseline interferometry) astrometry results spanning approximately 2.5 years for PSR J1012+5307, obtained with the Very Long Baseline Array as part of the MSPSRPI project. These provide the first proper motion and absolute position for PSR J1012+5307 measured in a quasi-inertial reference frame. From the VLBI results, we measure a distance of 0.83−0.02+0.060.83^{+0.06}_{-0.02}kpc (all the estimates presented in the abstract are at 68% confidence) for PSR J1012+5307, which is the most precise obtained to date. Using the new distance, we improve the uncertainty of measurements of the unmodeled contributions to orbital period decay, which, combined with three other pulsars, places new constraints on the coupling constant for dipole gravitational radiation κD=(−1.7±1.7)×10−4\kappa_D=(-1.7\pm1.7)\times 10^{-4} and the fractional time derivative of Newton's gravitational constant G˙/G=−1.8 −4.7 +5.6×10−13 yr−1\dot{G}/G = -1.8^{\,+5.6}_{\,-4.7}\times 10^{-13}\,{\rm yr^{-1}} in the local universe. As the uncertainties of the observed decays of orbital period for the four leading pulsar-WD systems become negligible in ≈10\approx10 years, the uncertainties for G˙/G\dot{G}/G and κD\kappa_D will be improved to ≤1.5×10−13 yr−1\leq1.5\times10^{-13}\,{\rm yr^{-1}} and ≤1.0×10−4\leq1.0\times10^{-4}, respectively, predominantly limited by the distance uncertainties.Comment: published in ApJ (2020ApJ...896...85D

    Glutamate Release in the Nucleus Accumbens Core Is Necessary for Heroin Seeking

    Long-term changes in glutamate transmission in the nucleus accumbens core (NAcore) contribute to the reinstatement of drug seeking after extinction of cocaine self-administration. Whether similar adaptations in glutamate transmission occur during heroin and cue-induced reinstatement of heroin seeking is unknown. After 2 weeks of heroin self-administration and 2 weeks of subsequent extinction training, heroin seeking was induced by a noncontingent injection of heroin or by presentation of light/tone cues previously paired with heroin infusions. Microdialysis was conducted in the NAcore during reinstatement of heroin seeking in animals extinguished from heroin self-administration or in subjects receiving parallel (yoked) noncontingent saline or heroin. Reinstatement by either heroin or cue increased extracellular glutamate in the NAcore in the self-administration group, but no increase was elicited during heroin-induced reinstatement in the yoked control groups. The increase in glutamate during heroin-induced drug seeking was abolished by inhibiting synaptic transmission in the NAcore with tetrodotoxin or by inhibiting glutamatergic afferents to the NAcore from the prelimbic cortex. Supporting critical involvement of glutamate release, heroin seeking induced by cue or heroin was blocked by inhibiting AMPA/kainite glutamate receptors in the NAcore. Interestingly, although a heroin-priming injection increased dopamine equally in animals trained to self-administer heroin and in yoked-saline subjects, inhibition of dopamine receptors in the NAcore also blocked heroin- and cue-induced drug seeking. Together, these findings show that recruitment of the glutamatergic projection from the prelimbic cortex to NAcore is necessary to initiate the reinstatement of heroin seeking

    Space shuttle, the most complex transportation system designed to date, illustrates the requirement for an analysis approach that considers all major disciplines simultaneously. Its unique cross coupling and high sensitivity to aerodynamic uncertainties and high performance requirements dictated a less conservative approach than those taken in programs. Analyses performed for the space shuttle and certain payloads, Space Telescope and Spacelab, are used a examples. These illustrate the requirements for system analysis approaches and criteria, including dynamic modeling requirements, test requirements control requirements and the resulting design verification approaches. A survey of the problem, potential approaches available as solutions, implications for future systems, and projected technology development areas are addressed

    Development of a Hard X-Ray Polarimeter for Astrophysics

    We have been developing a Compton scatter polarimeter for measuring the linear polarization of hard X-rays (100-300 keV) from astrophysical sources. A laboratory prototype polarimeter has been used to successfully demonstrate the reliability of our Monte Carlo simulation code and to demonstrate our ability to generate a polarized photon source in the lab. Our design concept places a self-containedpolarimeter module on the front-end of a a 5-inch position sensitive PMT (PSPMT). We are currently working on the fabrication of a science model based on this PSPMT concept. Although the emphasis of our development effort is towards measuring hard X-rays from solar flares, our design has the advantage that it is sensitive over a rather large field-of-view (\u3e1 steradian), a feature that makes it especially attractive for γ-ray burst studie
